Scientific Background

CD244 is a heterophilic receptor of the signaling lymphocytic activation molecule (SLAM) family, with its ligand being CD48. Triggered by homo- or heterotypic cell-cell interactions, SLAM receptors modulate the activation and differentiation of various immune cells, playing a role in regulating both innate and adaptive immune responses. Its activity is controlled by the presence or absence of small cytoplasmic adapter proteins, SH2D1A/SAP and/or SH2D1B/EAT-2. Functioning as an activating natural killer (NK) cell receptor, its activation involves association with SH2D1A and FYN, with downstream signaling primarily mediated by VAV1, and to a lesser extent, INPP5D/SHIP1 and CBL. Signal attenuation in the absence of SH2D1A is proposed to rely on INPP5D and, to a lesser degree, PTPN6/SHP-1 and PTPN11/SHP-2. CD244 stimulates NK cell cytotoxicity, IFN-gamma production, and granule exocytosis. Optimal NK cell expansion and activation appear dependent on CD244 engagement with CD48 expressed on neighboring NK cells (by similarity). It acts as a costimulator in NK activation by enhancing signals from other NK receptors such as NCR3 and NCR1 (by similarity). At early stages of NK cell differentiation, it may function as an inhibitory receptor, potentially ensuring self-tolerance of developing NK cells. CD244 is involved in regulating CD8(+) T-cell proliferation, where its expression on activated T-cells and binding to CD48 provides a costimulatory-like function for neighboring T-cells (by similarity). It also inhibits inflammatory responses in dendritic cells (DCs) (by similarity).
