Scientific Background
Activin Receptor Type IIB (ACVR2B) is a transmembrane serine/threonine kinase that forms an activin receptor complex with type I receptors (ACVR1, ACVR1B or ACVR1C). Functioning as the primary receptor for activin-A/INHBA, activin-B/INHBB and inhibin-A/INHA-INHBA, it regulates diverse processes including neuronal differentiation/survival, hair follicle cycling, pituitary FSH production, wound healing, extracellular matrix formation, immunosuppression and carcinogenesis (by similar: potential paracrine/autocrine roles in ovarian folliculogenesis). Upon activation, ACVR2B phosphorylates type I receptors to initiate SMAD2/SMAD3-SMAD4-mediated transcriptional regulation. Its signaling can be inhibited by SMAD7 (recruited via FKBP1A) or inhibin-B (bound through IGSF1 coreceptor).
