Scientific Background

Alpha-synuclein is a member of the synuclein family, which also includes beta- and gamma-synuclein. Alpha-synuclein is a neuronal protein that plays many roles in synaptic activity such as membrane trafficking of synaptic vesicles and neurotransmitter release. Multiplications in SNCA have been implicated in genetic forms of Parkinson disease and pathological post-translational modifications of alpha-synuclein are a hallmark of Parkinson’s disease. These modifications include phosphorylation, nitration, acetylation, truncation, and aggregation. C-terminal truncations of alpha-synuclein lead to increased aggregation. Evidence for the 1-122 truncation in brains of patients with Parkinson’s disease and Lewy Body Dementia (LBD).
