BRAT1 is a component of a multiprotein complex required for the assembly of the RNA endonuclease module of the integrator complex. It associates with INTS9 and INTS11 in the cytoplasm, blocking the active site of INTS11 to inhibit its endonuclease activity prior to full integrator complex formation. After dissociation of WDR73, BRAT1 facilitates nuclear import of the INTS9-INTS11 heterodimer. In the nucleus, INTS4 is integrated into the heterodimer, and BRAT1 is released from the mature RNA endonuclease module by inositol hexakisphosphate (InsP6). BRAT1 also participates in DNA damage response by activating kinases ATM, SMC1A, and PRKDC through phosphorylation modulation following ionizing radiation (IR) stress. Additionally, it regulates mitochondrial function and cell proliferation, maintains protein stability of MTOR and related proteins, and promotes growth factor-dependent cell cycle progression.
