Scientific Background

BRG1 is an ATPase involved in transcriptional activation and repression of select genes through chromatin remodeling (alteration of DNA-nucleosome topology). It is a component of SWI/SNF chromatin remodeling complexes, which modify chromatin structure by altering DNA-histone contacts within nucleosomes in an ATP-dependent manner. BRG1 is also part of the CREST-BRG1 complex, which regulates promoter activation by coordinating calcium-dependent release of a repressor complex and recruitment of an activator complex. In resting neurons, transcription of the c-FOS promoter is inhibited by SMARCA4-dependent recruitment of a phospho-RB1-HDAC repressor complex. Upon calcium influx, RB1 is dephosphorylated by calcineurin, leading to repressor complex release and increased recruitment of CREBBP to the promoter via a CREST-dependent mechanism, resulting in transcriptional activation. The CREST-BRG1 complex also binds the NR2B promoter, where activity-dependent induction of NR2B expression involves HDAC1 release and CREBBP recruitment (By similarity). BRG1 belongs to the neural progenitor-specific chromatin remodeling complex (npBAF complex) and the neuron-specific chromatin remodeling complex (nBAF complex). During neural development, a switch occurs from a stem/progenitor to a postmitotic chromatin remodeling mechanism as neurons exit the cell cycle. The npBAF complex contains ACTL6A/BAF53A and PHF10/BAF45A, while the nBAF complex contains homologous alternative subunits ACTL6B/BAF53B and DPF1/BAF45B or DPF3/BAF45C. The npBAF complex is essential for neural stem cell self-renewal/proliferation, whereas the nBAF complex, along with CREST, regulates genes critical for dendrite growth (By similarity). Additionally, BRG1 acts as a corepressor of ZEB1 to regulate E-cadherin transcription and is required for ZEB1-induced epithelial-mesenchymal transition (EMT). It binds enhancers between DLX5 and DLX6 via DLX1, stabilized by the lncRNA Evf2 (By similarity). BRG1 also binds RNA promiscuously (By similarity) and is involved in regulating thermogenic gene expression in brown adipose tissue (By similarity).
