Scientific Background
CD229 is a self-ligand receptor of the signaling lymphocytic activation molecule (SLAM) family. Triggered by homo- or heterotypic cell-cell interactions, SLAM receptors modulate the activation and differentiation of diverse immune cells, thereby regulating and interconnecting innate and adaptive immune responses. Its activity is controlled by the presence or absence of small cytoplasmic adapter proteins SH2D1A/SAP and/or SH2D1B/EAT-2. CD229 may participate in adhesion between T lymphocytes and accessory cells via homophilic interaction, promoting T-cell differentiation into a Th17 phenotype with increased IL-17 secretion (this costimulatory activity requires SH2D1A). It also recruits RORC to the IL-17 promoter and may act as a negative regulator of immune responses to maintain peripheral tolerance by suppressing autoantibody responses and IFN-gamma secretion by CD4(+) T-cells. By similarity, it may negatively regulate thymic innate CD8(+) T-cell populations and invariant natural killer T (iNKT) cell development.
