Scientific Background
CD31 is a cell adhesion molecule essential for leukocyte transendothelial migration (TEM) under inflammatory conditions. Its Tyr-690 residue critically regulates TEM by facilitating PECAM1 trafficking to/from the lateral border recycling compartment (LBRC) and directing LBRC membrane targeting around migrating leukocytes. CD31 mediates endothelial cell-cell adhesion via homophilic interactions and promotes neutrophil TEM through heterophilic binding with CD177. Homophilic ligation transmits a detachment signal to prevent phagocytosis of viable leukocytes by macrophages, while tethering apoptotic leukocytes to phagocytes (the detachment signal is disabled in apoptosis). It modulates BDKRB2 activation and regulates bradykinin/hyperosmotic shock-induced ERK1/2 signaling in endothelial cells (By similarity: may confer atherosclerosis susceptibility but lacks anti-apoptotic protection).
