Scientific Background
CDC34 is an E2 ubiquitin-conjugating enzyme that accepts ubiquitin from an E1 ubiquitin-activating protein and catalyzes its covalent attachment to other proteins via an E3 ubiquitin-protein ligase complex. In vitro, it mediates 'Lys-48'-linked polyubiquitination. CDC34 cooperates with the E2 UBCH5C and the SCF(FBXW11) E3 ligase complex to polyubiquitinate NFKBIA, leading to its proteasomal degradation. It also works with the SCF(SKP2) E3 ligase complex to regulate cell proliferation by ubiquitinating and degrading MYBL2 and KIP1. Additionally, CDC34 is involved in the degradation of CREM isoform ICERIIgamma and ATF5, abolishing their repression of cAMP-induced transcription during meiotic and mitotic cycles. It regulates the G2/M phase of the cell cycle by targeting WEE1 kinase for ubiquitination and degradation and participates in beta-catenin degradation. CDC34 is also a target of the human herpes virus 1 protein ICP0, modulating its interaction with proteasomes.
