Scientific Background
CLEC2 is a C-type lectin-like receptor that functions as a platelet receptor for the lymphatic endothelial marker PDPN. Upon ligand activation, it signals via sequential activation of SRC and SYK tyrosine kinases, leading to PLCG2 activation. In microbial infections, CLEC2 acts as a receptor for the snake venom protein rhodocytin, triggering tyrosine phosphorylation and SYK binding, which initiates downstream phosphorylation events and PLCG2 activation. Additionally, CLEC2 serves as an attachment factor for Human immunodeficiency virus type 1 (HIV-1), facilitating its capture by platelets.
