Scientific Background

Calcium-responsive transactivator (CREST), also known as SS18L1, is a calcium-dependent neuronal BAF family member expressed in the developing brain (1-4). CREST recruits CBP to nuclear bodies in order to regulate cortical and hippocampal dendrite development (1,5). The activity of CREST is aided by the ATPase of the BAF complex, Brg1, which upon calcium influx, will release a phospho-Rb/HDAC1 repressive complex from neuronal genes (4). CREST mutations have been described in ALS. The protein contains a prion-like domain, associates with the ALS protein FUS, forms aggregates, and is included in paraspeckles (6-8).
