Scientific Background

CaMKII β is a calcium/calmodulin-dependent protein kinase that operates autonomously following Ca2+/calmodulin binding and autophosphorylation, playing roles in dendritic spine/synapse formation, neuronal plasticity, and regulation of sarcoplasmic reticulum Ca2+ transport in skeletal muscle. In neurons, it structurally reorganizes the actin cytoskeleton during plasticity by kinase-independent actin filament bundling, enabling proper targeting of CaMK2A to promote synaptic plasticity underlying long-term potentiation (LTP) and hippocampus-dependent learning. It facilitates dendritic arborization in developing hippocampal neurons and remodeling in mature neurons, while also regulating neuronal migration. In skeletal muscle, it modulates function during exercise: regulating SR Ca2+ transport in slow-twitch fibers and controlling Ca2+ release via phosphorylation of triadin and phospholamban in fast-twitch fibers. Upon IFN-γ stimulation, it phosphorylates STAT1 to activate JAK-STAT signaling (By similarity). Under ER stress, it phosphorylates RETREG1 at Ser-151 to enhance its oligomerization and reticulophagy activity.
