Scientific Background

Cannabinoid Receptor I (CNR1) is a G-protein coupled receptor that binds endogenous cannabinoids (eCBs) including N-arachidonoylethanolamide (anandamide or AEA) and 2-arachidonoylglycerol (2-AG), as well as phytocannabinoids like delta(9)-tetrahydrocannabinol (THC). It mediates diverse cannabinoid-induced effects such as food intake regulation, memory impairment, gastrointestinal motility, catalepsy, locomotor activity, anxiety modulation, and chronic pain management, primarily through cyclic AMP reduction. In the hypothalamus, CNR1 exhibits dose-dependent dual effects on mitochondrial respiration: enhancing respiration at low doses while suppressing it at high doses via G-protein alpha-i activation, soluble adenylate cyclase inhibition, and PKA-dependent phosphorylation blockade of mitochondrial electron transport chain components (e.g., NDUFS2). CNR1 also inhibits leptin-induced ROS formation and upregulates SREBF1/FASN expression. By stimulating orexigenic beta-endorphin (but not alpha-MSH) release from POMC neurons, it promotes feeding behavior. Hippocampal CNR1 regulates cannabinoid-dependent cellular respiration and participates in depolarization-induced suppression of inhibition (DSI), where postsynaptic depolarization triggers retrograde eCB signaling to transiently reduce GABAergic transmission (By similarity). In vascular smooth muscle cells, CNR1 constitutively inhibits voltage-gated Ca2+ channels, inducing vasodilation (By similarity). Adipose CNR1 signaling elevates SREBF1/ACACA/FASN expression (By similarity), while hepatic activation increases lipogenesis and decreases fatty acid oxidation via SREBF1/GCK/ACACA/ACACB/FASN upregulation. It may impair energy expenditure in striated muscles by downregulating mitochondrial DLD and glycolytic enzymes (By similarity). In macrophages, anandamide-activated CNR1 triggers NLRP3 inflammasome-mediated IL1B/IL18 secretion, potentially contributing to type-2 diabetes progression (By similarity). Notably, CNR1 binds both 2-AG and anandamide, though 2-AG acts as an inverse agonist in certain isoforms (e.g., isoform 2/3).
