Scientific Background

Chk2 is a serine/threonine-protein kinase essential for checkpoint-mediated cell cycle arrest, DNA repair activation, and apoptosis in response to DNA double-strand breaks. It phosphorylates CDC25A/B/C to inhibit their activity, blocking cell cycle progression, and enhances homologous recombination repair by phosphorylating BRCA2 to promote RAD51-chromatin association. Chk2 regulates apoptosis via phosphorylation of p53/TP53, MDM4, and PML, while activating DNA repair and pro-apoptotic gene transcription through FOXM1 and E2F1. Its tumor suppressor role extends to mitotic spindle assembly via BRCA1 phosphorylation. Under oxidative stress, Chk2 phosphorylates TRIM32 to ubiquitinate ATG7, promoting autophagosome assembly (by similar: during microbial infection, it phosphorylates HHV-1 ICP0 to activate its SUMO-targeted ubiquitin ligase activity).
