Cleaved Caspase-3 (Asp175) (D3E9) Rabbit Monoclonal Antibody (SignalFlexTMAlexa Fluor®532 Conjugate)#17926,Cell Signaling Technology (CST),17926

Cleaved Caspase-3 (Asp175) (D3E9) Rabbit mAb (SignalFlexTM Alexa Fluor®532 Conjugate) recognizes endogenous levels of caspase-3 protein only when cleaved at Asp175. This antibody is preferred for immunofluorescence, flow cytometry, and immunohistochemistry applications. This antibody detects non-specific caspase substrates by western blot. Non-specific labeling may be observed by immunofluorescence in specific sub-types of healthy cells in fixed-frozen tissues (e.g. pancreatic alpha-cells). Nuclear background may be observed in rat and monkey samples.

FormulationSignalFlexTM conjugates are produced using highly validated Cell Signaling Technology®primary antibodies and conjugation methods that have been rigorously tested, ensuring high-quality conjugates and lot-to-lot consistency. These conjugates are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity. However, they are not tested on specific assays.Optimal dilutions/concentrations should be determined by the end user. When performing flow cytometry, we recommend using an isotype control conjugate at the same concentration as the antibody conjugate.
StorageSupplied in PBS (pH 7.2), less than 0.1% sodium azide, and 2 mg/mL BSA. Store at 4°C.Do not aliquot the antibody. Protect from light. Do not freeze.

Scientific Background

Caspase-3 (CPP-32, Apopain, Yama, SCA-1) is a critical executioner of apoptosis, as it is either partially or totally responsible for the proteolytic cleavage of many key proteins, such as the nuclear enzyme poly (ADP-ribose) polymerase (PARP) (1). Activation of caspase-3 requires proteolytic processing of its inactive zymogen into activated p17 and p12 fragments. Cleavage of caspase-3 requires the aspartic acid residue at the P1 position (2).Fernandes-Alnemri, T. et al. (1994)J Biol Chem269, 30761-4.Nicholson, D.W. et al.
