Scientific Background

CHOP is a multifunctional transcription factor primarily involved in the endoplasmic reticulum (ER) stress response. It plays a crucial role in various cellular stress responses and induces cell cycle arrest and apoptosis under ER stress conditions. CHOP exhibits dual functionality as both an inhibitor of CCAAT/enhancer-binding protein (C/EBP) and an activator of other genes (By similarity). It acts as a dominant-negative regulator of C/EBP by dimerizing with its family members, impairing their binding to promoter regions and suppressing C/EBP-regulated gene expression (By similarity). CHOP positively regulates the transcription of TRIB3, IL6, IL8, IL23, TNFRSF10B/DR5, PPP1R15A/GADD34, BBC3/PUMA, BCL2L11/BIM, and ERO1L, while negatively regulating BCL2, MYOD1, ASNS, HAMP, and PPARG expression. In collaboration with ATF4, CHOP mediates ER stress-induced cell death by promoting genes related to amino acid metabolism, mRNA translation, and the unfolded protein response (UPR) (By similarity). Additionally, CHOP inhibits the canonical Wnt signaling pathway by binding to TCF7L2/TCF4 and participates in inflammatory response regulation by inducing caspase-11 (CASP4/CASP11) and caspase-1 (CASP1) activation, which process pro-IL1B into mature IL1B (By similarity). It also serves as a key regulator of postnatal neovascularization through endothelial nitric oxide synthase (NOS3)-related signaling (By similarity).
