Scientific Background
7-dehydrocholesterol reductase (DHCR7) catalyzes the conversion of 7-dehydrocholesterol (7-DHC) to cholesterol, the last step of cholesterol biosynthesis. Studies show that DHCR7 expression is downregulated in macrophages upon viral infection. DHCR7 deficiency activates the PI3K-AKT3 pathway. AKT3 phosphorylates interferon regulatory factor 3 (IRF3) at Ser385, leading to IRF3 activation and increased type I interferon (IFN-I) production in macrophages (1). In addition, DHCR7 has proferroptotic activity. Its substrate, 7-DHC, suppresses ferroptosis by inhibiting phospholipid peroxidation.
