Scientific Background

DMRT1 is a transcription factor that plays a key role in male sex determination and differentiation by controlling testis development and male germ cell proliferation. It functions centrally in spermatogonia by inhibiting meiosis in undifferentiated spermatogonia and promoting mitosis, thereby driving spermatogonial development and enabling continuous sperm production. DMRT1 acts as both a transcriptional repressor and activator: it prevents meiosis by restricting retinoic acid (RA)-dependent transcription and repressing STRA8 expression, while activating spermatogonial differentiation genes like SOHLH1 to promote development. It also maintains postnatal sex determination by sustaining testis identity and preventing feminization through repression of female-promoting genes (e.g., FOXL2) and activation of male-specific genes. DMRT1 may function as a tumor suppressor. By similarity, it might play a minor role in oogenesis.
