Scientific Background

DNPH1 participates in a nucleotide salvage pathway that removes epigenetically modified 5-hydroxymethyl-dCMP (hmdCMP) via a two-step mechanism: deamination to cytotoxic 5-hydroxymethyl-dUMP (hmdUMP), followed by hydrolysis of hmdUMP's N-glycosidic bond into 5-hydroxymethyluracil (hmU) and 2-deoxy-D-ribose 5-phosphate (deoxyribosephosphate). This process prevents genomic integration of the cytotoxic nucleotide.
