Scientific Background

E2F6 is an inhibitor of E2F-dependent transcription, functioning by cooperatively binding DNA with DP proteins through the E2 recognition site 5'-TTTC[CG]CGC-3', with a preference for 5'-TTTCCCGC-3'. Lacking transcriptional activation and pocket protein binding domains, it regulates a subset of E2F-dependent genes essential for cell cycle entry but not normal progression. It may silence meiosis-specific genes (e.g., SLC25A31/ANT4, by similarity) via recruitment of a chromatin remodeling complex with histone H3-K9 methyltransferase activity. Overexpression delays S-phase exit.
