FAM111A is a single-stranded DNA-binding serine protease that mediates the proteolytic cleavage of covalent DNA-protein cross-links (DPCs) during DNA synthesis, playing a key role in maintaining genomic integrity. DPCs are highly toxic DNA lesions that interfere with essential chromatin transactions (e.g., replication and transcription) and are induced by reactive agents like UV light or formaldehyde. FAM111A prevents replication fork stalling by removing DPCs, such as covalently trapped topoisomerase 1 (TOP1) adducts or PARP inhibitor-trapped PARP1-DNA complexes. It also promotes PCNA loading at replication sites and facilitates S-phase entry and DNA synthesis. Additionally, FAM111A acts as a restriction factor for viruses like SV40 polyomavirus and vaccinia virus by disrupting the nuclear pore complex (NPC) via its protease activity during viral replication; by similar, it interacts with vaccinia virus DNA-binding protein OPG079 in the cytoplasm and promotes its degradation through autophagy (independent of protease activity).
