Scientific Background

HDAC2 is a histone deacetylase that catalyzes the deacetylation of lysine residues on the N-terminal regions of core histones (H2A, H2B, H3, and H4). Histone deacetylation serves as an epigenetic repression marker and plays a key role in transcriptional regulation, cell cycle progression, and developmental events (by similar). It functions by forming large multiprotein complexes (by similar) and associates with MAD, SIN3, YY1, and N-COR to establish transcriptional repressor complexes. As part of the RCOR/GFI/KDM1A/HDAC complex, HDAC2 suppresses genes involved in multilineage blood cell development via histone deacetylase recruitment (by similar). It is also a component of the histone deacetylase NuRD complex, which participates in chromatin remodeling. Additionally, HDAC2 contributes to the SIN3B complex, repressing transcription and counteracting EP300's histone acetyltransferase activity by recognizing H3K27ac marks via PHF12 and leveraging its deacetylase function. Beyond histones, HDAC2 deacetylates non-histone targets like TSHZ3, modulating its transcriptional repressor activity. It may also be involved in CRY1-mediated transcriptional repression of circadian genes such as PER1 through histone deacetylation (by similar). Furthermore, HDAC2 exhibits protein-lysine deacylase activity, removing crotonyl, lactyl, and 2-hydroxyisobutyryl groups from lysine residues, leading to protein decrotonylation, delactylation, and de-2-hydroxyisobutyrylation.
