Scientific Background

JAK2 is a non-receptor tyrosine kinase involved in diverse processes including cell growth, development, differentiation, and histone modifications, playing essential roles in both innate and adaptive immunity signaling. In the cytoplasm, it mediates signal transduction by associating with type I receptors (e.g., GHR, PRLR, LEPR, EPOR, MPL/TPOR) or type II receptors (e.g., IFN-α/β/γ and interleukin receptors). Upon ligand binding, JAK2 phosphorylates receptor cytoplasmic tails to create docking sites for STAT proteins, which it further phosphorylates to enable their nuclear translocation and transcriptional activation. For instance, during erythropoiesis, EPO stimulation triggers JAK2 autophosphorylation and EPOR association, leading to STAT5 activation and erythropoiesis-related gene expression. JAK2 also participates in cellular retinol-induced STAT5 activation, angiotensin-2-driven ARHGEF1 phosphorylation, cell cycle regulation (via CDKN1B phosphorylation), and TEC-coordinated FOS transcription. In the nucleus, it phosphorylates histone H3 at Tyr-41 (H3Y41ph), excluding CBX5 (HP1α) from chromatin, and enhances KCNA3 potassium channel activity.
