Scientific Background
KDM1B is a histone demethylase that specifically removes mono- and di-methylation from 'Lys-4' of histone H3 (H3K4me1/me2), an epigenetic mark for transcriptional activation, thus functioning as a transcriptional corepressor. It catalyzes demethylation via FAD-dependent oxidation to generate an imine intermediate followed by hydrolysis. During oogenesis, KDM1B is required for de novo DNA methylation of a subset of imprinted genes. Notably, KDM1B shows no activity toward H3K4me3 or other histone methylation sites (e.g., H3K9/27/36me, H4K20me). Alone, it cannot demethylate nucleosomal H3K4me but requires GLYR1 to form a multifunctional enzyme complex that modifies transcribed chromatin and facilitates Pol II transcription through nucleosomes.
