EZH2, a Polycomb group (PcG) protein, acts as the catalytic subunit of the PRC2/EED-EZH2 complex, contributing to histone methylation and subsequent transcriptional repression. This complex mono-, di-, and trimethylates 'Lys-27' (H3K27) of histone H3, generating H3K27me1, H3K27me2, and H3K27me3, respectively. EZH2 exhibits a preference for substrates with less methylation and is pivotal for forming H3K27me3 in embryonic stem cells, playing a crucial role in maintaining stem cell identity and facilitating proper differentiation.
