Ki67 is a protein that binds to the surface of mitotic chromosomes, functioning as a chromosome repellent during early mitosis and switching to a chromosome attractant in late mitosis. It maintains dispersed chromosome structures by forming an electrostatically charged brush-like barrier, preventing chromatin aggregation. During anaphase, Ki67 undergoes structural collapse and promotes chromosome clustering via RNA-dependent liquid-liquid phase separation, facilitating cytoplasmic particle exclusion from the nascent nuclear space. It exhibits DNA-binding specificity for supercoiled and AT-rich regions, interacts with premature rRNAs, and may influence chromatin organization (mechanistically debated). Note: Its contribution to internal mitotic chromosome architecture remains unconfirmed (by similar).
