Scientific Background
LC3B is a ubiquitin-like modifier involved in the formation of autophagosomal vacuoles (autophagosomes). It plays a role in mitophagy, regulating mitochondrial quantity and quality by eliminating mitochondria to a basal level to meet cellular energy demands and prevent excessive ROS production. Under cellular stress and mitochondrial fission, LC3B binds C-18 ceramides and anchors autophagolysosomes to outer mitochondrial membranes to remove damaged mitochondria. Unlike LC3s, which contribute to phagophore membrane elongation, the GABARAP/GATE-16 subfamily is essential for later stages of autophagosome maturation. LC3B also promotes primary ciliogenesis by removing OFD1 from centriolar satellites via autophagy. Through interaction with the reticulophagy receptor TEX264, LC3B participates in remodeling endoplasmic reticulum subdomains into autophagosomes during nutrient stress, which then fuse with lysosomes for endoplasmic reticulum turnover. Under nutrient stress, LC3B directly recruits cofactor JMY to phagophore membrane surfaces, enhancing JMY's actin nucleation activity and autophagosome biogenesis during autophagy.
