LIN28 is an RNA-binding protein that inhibits the processing of pre-let-7 miRNAs and regulates the translation of mRNAs involved in developmental timing, pluripotency, and metabolism (e.g., binding IGF2 mRNA, MYOD1 mRNA). It recognizes G-quartet (G4) structural features in miRNA/mRNA targets, acting as a 'translational enhancer' to drive specific mRNAs to polysomes and boost protein synthesis. By associating with the translational machinery, LIN28 increases initiation events per mRNA and stabilizes mRNAs indirectly. It recruits TUT4/TUT7 uridylyltransferases to uridylate pre-miRNAs (e.g., let-7, miR-143), leading to their Dicer-blocked degradation, thereby maintaining embryonic stem cell pluripotency (similar mechanisms apply to miR107, miR-200c). Localized near the ER, LIN28 suppresses translation of ER-targeted mRNAs (e.g., transmembrane/secretory proteins) but enhances translation of metabolic enzymes (e.g., PFKP, PDHA1, SDHA), promoting glycolysis and oxidative phosphorylation (by similarity suggests a role in adult tissue repair).
