LRP2 is a multiligand endocytic receptor that collaborates with CUBN to mediate endocytosis of high-density lipoproteins (HDL). It facilitates receptor-mediated uptake of polybasic drugs such as aprotinin, aminoglycosides, and polymyxin B (By similarity). In the kidney, LRP2 mediates tubular uptake and clearance of leptin and enables its transport across the blood-brain barrier via choroid plexus epithelium endocytosis (By similarity). Neuronal leptin endocytosis by LRP2 is essential for hypothalamic leptin signaling and regulation of feeding and body weight. The receptor also mediates endocytosis and lysosomal degradation of CST3 in proximal tubule cells and, in complex with GC/DBP, facilitates renal uptake of 25-hydroxyvitamin D3. Additionally, LRP2 participates in renal uptake of metallothionein-bound heavy metals (By similarity) and cooperates with CUBN to reabsorb myoglobin (By similarity). It contributes to kidney selenium homeostasis by endocytosing SEPP1 and plays a role in renal uptake of BIRC5/survivin, which may support kidney integrity. LRP2 also mediates uptake of MMP2-TIMP1 complexes. During embryogenesis, LRP2 degrades BMP4 in neuroepithelia, ensuring proper SHH ventral neural tube localization and telencephalic patterning. In adult brains, it suppresses BMP signaling in the subependymal zone to sustain neurogenesis. Optic nerve development relies on LRP2 for SHH-driven oligodendrocyte precursor migration/proliferation, while retinal SHH clearance by LRP2 maintains progenitor quiescence. The receptor promotes reproductive organ development by internalizing SHBG-bound sex hormones and endocytoses angiotensins (By similarity). It also binds β-amyloid 40-CLU/APOJ complexes for lysosomal degradation (By similarity). LRP2 is vital for embryonic heart development and normal hearing, potentially via inner ear estrogen interactions.
