Scientific Background
The M-CSF Receptor is a tyrosine-protein kinase that functions as a cell-surface receptor for CSF1 and IL34, playing a pivotal role in regulating the survival, proliferation, and differentiation of hematopoietic precursor cells, particularly mononuclear phagocytes like macrophages and monocytes. It promotes the release of pro-inflammatory chemokines in response to IL34 and CSF1, contributing to innate immunity and inflammatory processes. Additionally, it is crucial for osteoclast proliferation, bone resorption, and normal bone/tooth development, as well as fertility and mammary gland development during pregnancy. The receptor facilitates actin cytoskeleton reorganization, membrane ruffle formation, cell adhesion, migration, and cancer cell invasion. Upon ligand binding, it activates signaling pathways such as ERK1/2 and JNK (By similarity), phosphorylates PIK3R1, PLCG2, GRB2, SLA2, and CBL, and subsequently triggers PKC family members (e.g., PRKCD) and the AKT1 pathway. It also mediates MAPK1/ERK2, MAPK3/ERK1, and SRC-family kinases (SRC, FYN, YES1) activation. Signaling occurs via direct interaction with phosphorylated tyrosines or adapters like GRB2, leading to STAT3/STAT5A/B activation and SHC1/INPP5D/SHIP-1 phosphorylation. Receptor signaling is downregulated by phosphatases (e.g., INPP5D/SHIP-1) and internalization. In the central nervous system, it may participate in microglia macrophage development (By similarity).
