MiTF is a transcription factor that serves as a master regulator of melanocyte survival, differentiation, and melanosome biogenesis. It binds to M-boxes (5'-TCATGTG-3') and E-boxes (5'-CACGTG-3') in the promoters of pigmentation genes like tyrosinase (TYR). Under nutrient-rich conditions, MTOR-mediated phosphorylation inactivates MiTF, while starvation or lysosomal stress triggers its dephosphorylation and transcriptional activation via MTOR inhibition. MiTF critically regulates melanocyte development by controlling TYR and TYRP1 expression and is involved in differentiating neural crest-derived melanocytes, mast cells, osteoclasts, and retinal pigment epithelium. by similar: MiTF's role is conserved across diverse cell-type differentiation processes.
