Scientific Background

Mitofusin 2 (Mfn2) is a mitochondrial membrane protein that regulates mitochondrial fusion, bioenergetics, and inter-organelle communication. It plays a vital role in maintaining mitochondrial integrity and function in neurons, which are highly dependent on energy metabolism. Loss or dysfunction of Mfn2 impairs mitochondrial dynamics, leading to fragmented mitochondria, disrupted membrane potential, and increased oxidative stress. These mitochondrial abnormalities are implicated in neurodegenerative diseases such as Charcot-Marie-Tooth disease type 2A, Alzheimer’s disease, and Parkinson’s disease. Mfn2 also modulates mitochondrial transport along axons and participates in mitophagy, the selective degradation of damaged mitochondria. Its role in regulating apoptosis and inflammation further underscores its importance in neuronal survival. Targeting Mfn2 to restore mitochondrial dynamics and function represents a promising strategy for mitigating neurodegenerative processes and enhancing neuronal resilience.
