Scientific Background
The deduced 1,203-amino acid protein contains an RNA recognition motif. RT-PCR ELISA detected moderate expression in all tissues and specific brain regions examined, with highest expression in cerebellum. Schneider et al. (2006) stated that the 1,727-amino acid TNRC6B protein contains a glycine/tryptophan (GW)-rich N-terminal domain, a central glutamine-rich region, and a C-terminal RNA recognition motif. TNRC6B copurified with a cytoplasmic HeLa cell protein complex containing AGO2 (EIF2C2), DICER, and MOV10. Epitope-tagged TNRC6B coimmunoprecipitated with tagged AGO1 (EIF2C1) and tagged AGO2. TNRC6B was required to mediate microRNA-guided mRNA cleavage in HeLa cell culture.
