NTH1 is a bifunctional DNA N-glycosylase with associated apurinic/apyrimidinic (AP) lyase function that initiates base excision repair (BER), the primary pathway for repairing oxidative DNA damage. Its DNA N-glycosylase activity cleaves the N-glycosidic bond to excise damaged bases, generating an AP site, while the AP-lyase activity cleaves the 3' phosphodiester bond via β-elimination. It primarily recognizes and repairs oxidative pyrimidine damage but also exhibits 8-oxo-7,8-dihydroguanine (8-oxoG) DNA glycosylase activity, preferentially targeting lesions opposite guanine residues. Notably, it processes nucleosome-embedded lesions without requiring or inducing nucleosome disruption.
