Nanog is a transcription regulator primarily involved in the proliferation and self-renewal of inner cell mass and embryonic stem (ES) cells. It imposes pluripotency on ES cells while preventing their differentiation into extraembryonic endoderm and trophectoderm lineages. By physically interacting with SMAD1, Nanog blocks bone morphogenetic protein-induced mesoderm differentiation of ES cells through interfering with coactivator recruitment to active SMAD transcriptional complexes. It functions as either a transcriptional activator or repressor, with optimal binding to the DNA consensus sequences 5'-TAAT[GT][GT]-3' or 5'-[CG][GA][CG]C[GC]ATTAN[GC]-3'. Nanog binds to the POU5F1/OCT4 promoter and autorepresses its own expression in differentiating ES cells via interaction with ZNF281/ZFP281, leading to NuRD complex recruitment. Overexpression of Nanog promotes S-phase entry and cell proliferation.
