Scientific Background

PAR4 is a pro-apoptotic protein that selectively induces apoptosis in cancer cells, sensitizing them to diverse apoptotic stimuli and causing tumor regression in animal models. It triggers apoptosis in certain cancer cells by activating the Fas pro-death pathway while simultaneously inhibiting NF-kappa-B transcriptional activity. PAR4 suppresses WT1-mediated transcriptional activation and enhances its repressive function, downregulating the anti-apoptotic protein BCL2 through interaction with WT1. Additionally, PAR4 appears to function as a transcriptional repressor itself and may directly regulate BACE1's cleavage activity of the amyloid precursor protein (APP).
