CD140a is a tyrosine-protein kinase that functions as a cell-surface receptor for PDGFA, PDGFB, and PDGFC, playing pivotal roles in embryonic development, cell proliferation, survival, and chemotaxis regulation. Its context-dependent actions include promoting or inhibiting cell proliferation/migration. It is essential for bone marrow-derived mesenchymal stem cell differentiation, embryonic skeleton development, cephalic closure, gastrointestinal mucosa formation, mesenchymal cell recruitment, and intestinal villi development. It contributes to wound healing via chemotaxis/migration and regulates platelet activation, granule secretion, and thrombin-induced aggregation. Ligand binding (PDGFA/PDGFB homo/heterodimers or PDGFC homodimers) triggers diverse signaling cascades modulated by PDGFRA/PDGFRB heterodimerization. It phosphorylates PIK3R1 (activating AKT1), PLCG1 (generating DAG/IP3 for Ca2+ mobilization and PKC activation), and PTPN11, while activating HRAS, MAPK1/ERK2/MAPK3/ERK1, and STAT1/STAT3/STAT5. Signaling is attenuated by phosphatase-mediated dephosphorylation and receptor internalization.
