PI3 Kinase Class 3 is the catalytic subunit of the PI3K complex, mediating the formation of phosphatidylinositol 3-phosphate. Different complex forms are involved in multiple membrane trafficking pathways: PI3KC3-C1 participates in the initiation of autophagosomes, while PI3KC3-C2 is involved in autophagosome maturation and endocytosis. As part of PI3KC3-C1, it promotes endoplasmic reticulum membrane curvature formation prior to vesicle budding. It also regulates degradative endocytic trafficking and is required for the abscission step in cytokinesis, likely in the context of PI3KC3-C2. By similarity, it is involved in the transport of lysosomal enzyme precursors to lysosomes and facilitates transport from early to late endosomes. In microbial infection, its kinase activity is essential for SARS-CoV-2 replication.
