Scientific Background

PKC delta, a calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase, plays dual roles in cell death and survival, acting as a pro-apoptotic protein during DNA damage-induced apoptosis and an anti-apoptotic protein during cytokine receptor-initiated cell death. It is implicated in tumor suppression, required for oxygen radical production by NADPH oxidase, and functions as a positive or negative regulator in platelet responses. PKC delta negatively regulates B cell proliferation and plays a crucial role in self-antigen-induced B cell tolerance induction. Under various cellular stresses, PKC delta activates signaling pathways leading to apoptosis, including the activation of death-promoting transcription factor BCLAF1/Btf, modulation of the CHUK/IKKA-p53 axis, and formation of a complex with ABL1. Moreover, it regulates cell cycle progression, mediates the inhibition of cell cycle progression at G1/S phase, and activates the G2/M DNA damage checkpoint. PKC delta is also involved in the control of cell cycle progression and protection of glioma cells from TNFSF10/TRAIL-induced apoptosis. Furthermore, it participates in antifungal immunity and regulates NADPH oxidase activity in neutrophils and eosinophils. PKC delta plays a critical role in platelet functions, acting as a negative regulator of filopodia formation and modulating granule secretion downstream of specific receptors. Its extensive phosphorylation targets, such as MUC1, ELAVL1, mitochondrial phospholipid scramblase 3 (PLSCR3), and SMPD1, underscore its versatility in modulating diverse cellular processes.
