Scientific Background

c-Yes is a non-receptor protein tyrosine kinase involved in regulating cell growth, survival, apoptosis, cell-cell adhesion, cytoskeleton remodeling, and differentiation. Stimulated by receptor tyrosine kinases (RTKs) such as EGFR, PDGFR, CSF1R, and FGFR, it is recruited to phosphorylated receptors, activating and phosphorylating downstream substrates. Upon EGFR activation, c-Yes promotes PARD3 phosphorylation to support epithelial tight junction assembly. It participates in phosphorylating junctional components like CTNND1 by stimulating FYN and FER tyrosine kinases at cell-cell contacts. During CXCL12-induced T-cell stimulation, c-Yes phosphorylates collapsin response mediator protein 2/DPYSL2, inducing T-cell migration. It also contributes to the CD95L/FASLG signaling pathway and mediates AKT-dependent cell migration. In cell cycle progression, c-Yes phosphorylates cyclin-dependent kinase 4/CDK4 to regulate the G1 phase and participates in G2/M progression and cytokinesis. Additionally, it catalyzes the phosphorylation of organic cation transporter OCT2, enhancing its transport activity.
