Phospho-TBK1/NAK (Ser172) (D52C2) Rabbit Monoclonal Antibody (SignalFlexTMAlexa Fluor®700 Conjugate)#23411,Cell Signaling Technology (CST),23411

Phospho-TBK1/NAK (Ser172) (D52C2) XP®Rabbit mAb (SignalFlexTM Alexa Fluor®700 Conjugate) detects endogenous levels of TBK1 only when phosphorylated at Ser172. This antibody may cross-react with phospho-IKKε. Phospho-TBK1/NAK (Ser172) (D52C2) XP®Rabbit mAb lacks sensitivity in fixed frozen mouse tissue by indirect immunofluorescent detection.

FormulationSignalFlexTM conjugates are produced using highly validated Cell Signaling Technology®primary antibodies and conjugation methods that have been rigorously tested, ensuring high-quality conjugates and lot-to-lot consistency. These conjugates are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity. However, they are not tested on specific assays.Optimal dilutions/concentrations should be determined by the end user. When performing flow cytometry, we recommend using an isotype control conjugate at the same concentration as the antibody conjugate.
StorageSupplied in PBS (pH 7.2), less than 0.1% sodium azide, and 2 mg/mL BSA. Store at 4°C.Do not aliquot the antibody. Protect from light. Do not freeze.

Scientific Background

TBK1 (TANK-binding kinase 1)/NAK (NF-κB activating kinase) is an IκB kinase (IKK)-activating kinase and can activate IKK through direct phosphorylation (1). TBK1 was identified through association with the TRAF binding protein, TANK, and found to function upstream of NIK and IKK in the activation of NF-κB (2). TBK1 induces IκB degradation and NF-κB activity through IKKβ. TBK1 may mediate IKK and NF-κB activation in response to growth factors that stimulate PKCε activity (1). TBK1 plays a pivotal role in the activation of IRF3 in the innate immune response (3).TBK1 is phosphorylated at Ser172 within its activation loop, which is necessary for its kinase activity including the downstream phosphorylation of IRF3 (4,5).Alternate NamesFLJ11330; FTDALS4; IIAE8; NAK; NF-kappa-B-activating kinase; NF-kB-activating kinase; Serine/threonine-protein kinase TBK1; T2K; TANK binding kinase 1; TANK-binding kinase 1; TBK1
