COUP-TFI (Chicken Ovalbumin Upstream Promoter-Transcription Factor I), also known as NR2F1, is a nuclear receptor and transcription factor that plays a pivotal role in neurodevelopment, cell differentiation, and gene regulation. It belongs to the steroid/thyroid hormone receptor superfamily and functions primarily by binding to specific DNA motifs (such as 5'-AGGTCA-3') to regulate gene expression either as a repressor or activator. COUP-TFI is essential for proper cortical development, neuronal migration, and axonal guidance, particularly in the central nervous system. Its expression is tightly regulated during embryogenesis and is critical for establishing regional identity in the developing brain. Dysregulation or mutations in the NR2F1 gene are associated with Bosch-Boonstra-Schaaf optic atrophy syndrome (BBSOAS), a rare neurodevelopmental disorder characterized by intellectual disability, optic nerve atrophy, and developmental delays. Additionally, aberrant COUP-TFI activity has been implicated in various cancers and metabolic disorders.
