For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with any of the following:- Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100- Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol- 100% ice-cold methanol onlyFor use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803). Fixation/permeabilization with either True-Phos™ Perm Buffer (Cat. No. 425401) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) is not recommended.This antibody does not cross react with mouse Calponin 1 in Western Blot.

Scientific Background

Calponin 1, encoded by the CNN1 gene, is an actin filament-associated regulatory protein found primarily in smooth muscle but also in non-muscle cells. Calponin 1 plays a significant role in regulating the contractility and structural integrity of the cytoskeleton by binding to actin and actin-interacting proteins, modulating their interplay. For example, by binding to actin and inhibiting myosin light chain kinase Calponin 1 reduces the phosphorylation of myosin light chains, preventing excessive contraction and ensuring appropriate smooth muscle response to physiological stimuli. Furthermore, Calponin 1 has been implicated in various physiological functions beyond muscle contraction including vascular tone and wound healing, making it a key player in maintaining homeostasis in various tissues.
