Scientific Background

Cyclophilin D is one of the three components of the permeability transition pore (PTP) localized in the mitochondrial matrix. The opening of the PTP is a key event in the initiation of cell death which leads to mitochondrial swelling, outer membrane rupture, and the release of apoptotic regulators such as cytochrome C. Cyclophilin D promotes mitochondrial permeability transition (mPT) in response to increased Ca2+level. Cyclophilin D deficient mice have shown desensitization to mTP and require twice as much Ca2+intake for the opening of the PTP compared to wild-type mice.
