DAP12, also known as TYROBP, is a transmembrane adapter protein expressed in various immune cells, including natural killer cells, monocytes, macrophages, dendritic cells, osteoclasts, and microglia. It plays a role in immune cell functions such as survival, proliferation, phagocytosis, chemotaxis, and cytokine production. DAP12 forms complexes with several immunoreceptors, including TREM2, to trigger intracellular signaling pathways. Regulation of DAP12 involves its interaction with proteins like RER1, which retains unassembled DAP12 in early secretory compartments, preventing its transport to the plasma membrane. Mutations in DAP12 and its associated receptors have been linked to neurodegenerative diseases, such as Nasu-Hakola disease and Alzheimer's disease. Additionally, DAP12 signaling can modulate immune responses, as seen in its paradoxical role in down-regulating cytokine production by plasmacytoid dendritic cells during viral infections.
