Scientific Background
ANTXR2, also known as anthrax toxin receptor 2, is a type I transmembrane protein with an extracellular von Willebrand factor A (vWA) domain, an immunoglobulin (Ig)-like domain, a single transmembrane helix and a cytoplasmic tail. It binds to theBacillus anthracistoxin subunit, protective antigen (PA), in a divalent cation-dependent manner. Upon binding of PA, multiple toxin-receptor complexes assemble in lipid rafts, which are then internalized. This results in the formation of a pore in the endosomal membrane, enabling the escape of other anthrax toxin subunits into the cytoplasm. Additionally, ANTXR2 interacts with laminin, and possibly collagen type IV, suggesting a role for this receptor in capillary formation and basement membrane assembly.
