Scientific Background

RB11 is a member of the small GTPase Rab family that serves as a key regulator of intracellular membrane trafficking, cycling between GDP-bound inactive and GTP-bound active states to control vesicle formation, movement, tethering, and fusion. It forms a functional Rab11/RAB11FIP3/dynein complex to direct the microtubule-dependent movement of peripheral sorting endosomes (SE) toward the centrosome, establishing the endosomal recycling compartment (ERC). As a major coordinator of membrane delivery during cytokinesis, RB11 collaborates with MYO5B and RAB8A in epithelial cell polarization and partners with Rabin8/RAB3IP, the exocyst complex, and other factors to enable PODXL transcytosis for apical membrane initiation site (AMIS) formation and lumenogenesis. It also mediates CFTR plasma membrane targeting, transferrin (TF) recycling, NPC1L1 transport to the plasma membrane, and Golgi-to-plasma membrane sorting/basolateral transport of CDH1. By similarity functions include FCGRT (monomeric IgG Fc receptor) recycling to basolateral membranes and potential roles in melanosome transport/release. RB11 promotes ciliogenesis initiation by assembling a ciliary targeting complex with ASAP1, Rabin8/RAB3IP, RAB11FIP3, and ARF4 to drive Rabin8 preciliary vesicle trafficking, though LPAR1-activated WDR44 phosphorylation disrupts this process. Recent studies reveal its involvement in a Rab8-Rab10-Rab11-endosomal export pathway for newly synthesized proteins via WDR44 interaction.
