Scientific Background

REST is a transcriptional repressor that binds to the neuron-restrictive silencer element (NRSE) and represses neuronal gene transcription in non-neuronal cells. It restricts neuronal gene expression by associating with two distinct corepressors, SIN3A and RCOR1, which recruit histone deacetylase to the promoters of REST-regulated genes. Additionally, REST mediates repression by recruiting the BHC complex to RE1/NRSE sites, acting as a chromatin modifier through histone deacetylation and demethylation (By similarity). In non-neural cells, REST represses SRRM4 expression to prevent neural-specific splicing events and the production of REST isoform 3 (By similarity). Its repressor activity may be inhibited by forming heterodimers with isoform 3, preventing binding to NRSE or corepressors and leading to derepression of target genes. REST also maintains repression of neuronal genes in neural stem cells and allows neuronal differentiation by dissociating from RE1/NRSE sites (By similarity). Furthermore, REST acts as a key repressor of gene expression in hypoxia by directly binding to RE1/NRSE sites on target gene promoters. In breast cancer cell lines, REST negatively regulates SRRM3 expression.
