Scientific Background

RFP Tag is an E3 ubiquitin-protein ligase that mediates ubiquitination of various substrates, playing roles in processes such as proliferation, innate immunity, apoptosis, immune response, or autophagy. It ubiquitinates PIK3C2B and inhibits its activity by forming 'Lys-48'-linked polyubiquitin chains, thereby suppressing CD4 T-cell activation (by similar). Additionally, RFP regulates retrograde transport by collaborating with MAGEL2 to form 'Lys-63'-linked polyubiquitin chains at 'Lys-220' of WASHC1, promoting endosomal F-actin assembly (by similar). RFP also exhibits transcriptional repressor activity in cooperation with EPC1 (by similar). In apoptosis regulation, RFP activates Jun N-terminal kinase and p38 kinase to induce apoptosis and enhances caspase-3-like activity independently of mitochondrial events (by similar). It may function in male germ cell development (by similar) and preferentially binds double-stranded DNA (by similar). RFP forms a complex with USP7 and ubiquitinates it, leading to deubiquitination of RIPK1 and positive regulation of TNF-alpha-induced apoptosis. Moreover, RFP, along with USP7 or PTPN11, inhibits the antiviral signaling pathway by promoting TBK1 ubiquitination and degradation (by similar).
