RING2 is an E3 ubiquitin-protein ligase that mediates monoubiquitination of 'Lys-119' on histone H2A (H2AK119Ub), playing a central role in the histone code and gene regulation. H2AK119Ub serves as a specific tag for epigenetic transcriptional repression and participates in X chromosome inactivation in female mammals (By similarity). It may be involved in initiating both imprinted and random X inactivation (By similarity). As a core component of the Polycomb group (PcG) multiprotein PRC1-like complex, it helps maintain the transcriptionally repressive state of genes, including Hox genes, during development (PubMed:16359901, PubMed:26151332). The PcG PRC1 complex remodels chromatin and modifies histones, leading to heritable changes in chromatin expressibility (PubMed:26151332). Its E3 ubiquitin-protein ligase activity is enhanced by BMI1/PCGF4 (PubMed:21772249). Within the PRC1 complex, RING2 acts as the primary E3 ubiquitin ligase for histone H2A, while RING1 may modulate RNF2/RING2 activity (Probable). Its association with chromosomal DNA is cell-cycle dependent. In resting B- and T-lymphocytes, interaction with AURKB inhibits its activity, sustaining transcription in these cells (By similarity). Additionally, it negatively regulates autophagy by ubiquitinating AMBRA1, leading to its degradation (By similarity).
