The recombinantAnti-β-Gal-mIgG2ae3is an Fc silent monoclonal antibody (mAb) that serve as a negative control antibodyin vivo. This control antibody targetsE. coli's β-galactosidase (β-Gal), an antigen that is not present in mice. Its engineered mouse IgG2a isotype contains three point mutations in the Fc region at positionsL234A, L235A, andP329G. These mutations, also referred to as LALA-PG, result in the complete loss of unwanted Fc-associated effector functions. Anti-β-Gal-mIgG2ae3 is expressed and produced in Chinese hamster ovary (CHO) cells, ensuring reliability and lot-to-lot reproducibility. Thereby, common hybridoma-related drawbacks such as the generation of non-relevant mAbs containing aberrant light chains are avoided. Due to the triple mutation LALA-PG, it displaysno ADCP(antibody-dependent cell-mediated phagocytosis),no ADCC(antibody-dependent cell-mediated cytotoxicity) andno CDC(complement-dependent cytotoxicity) effector functions. Thesequenceof Anti-β-Gal-mIgG2ae3 InvivoFit^TM is100% murine(constant and variable regions), as the original clone (clone T9C6) was raised in mice using a proprietary method. This feature allows for reduced immunogenicity and risks of fatal hypersensitivity reactions upon repeated mAb injections into mice [2].
